ABSTRACT

Objective To understanding the genetic subtype and its population and regional distribution of HIV-1 strains circulating in Yunnan province.Methods 788 plasma specimens collected in 2008-2009 from 15 distracts of Yunnan,were enrolled.Viral RNA were extracted and subjected to RT-PCR.1584 bp full length gag gene,3147 bp full length pol gene and 558 bp env (C2V3) fragment were amplified and directly sequenced.Full length gag and pol genes were connected together as a complete genetic region (location on HXB2:790-5096) for genotyping.Results Of the 788 plasma specimens,a total number of 1728 genomic sequences including 599 gag,564 pol and 525 env (C2V3) were successfully amplified and sequenced,with genotype of 617 samples identified.The subtypes of HIV-1 strains circulated in Yunnan were with the order of constituent ratio CRF08_BC ( 50.2% ),CRF01_AE (25.0%),unknown recombinant forms ( 10.2% ),CRF07_BC (9.2%),subtype C (2.9%) and subtype B (B') (2.4%).The distributions of subtypes showed significant regional differences and could be roughly divided into two forms:the CRF08_BC predominant areas represented by Lincang and Kunming,and the areas with CRF08_BC together with CRF01_AE coexistence,represented by Dehong and Xishuangbanna.The unknown recombinant forms accounted for more HIV infection in ethnic minorities (17.0%) than in ethnic Han (6.7%,P<0.01 ).The distribution of subtypes varied significantly in the two primary routes of transmission for those infected through heterosexual contact.CRF08_BC and CRF0 1_AE were the dominant subtypes,accounting for 52.7% and 29.1% respectively.However,in IDUs,CRF08_BC strains accounted for half of the infection,while only 4.5% of the infections were caused by CRF01_AE,CRF07_BC while the unique recombinant forms were responsible for 15.5% infections.Of the 63 unknown recombinant forms,most (74.6%) were B (B' ) recombinant with C,while 25% were mosaic B and/or C fragments on the bases of CRF01_AE genome.Conclusion The subtypes of HIV-1 strains circulated in Yunnan were complicated under the significant differences of regions,ethnics or routes of transmission.

ABSTRACT

<p><b>OBJECTIVE</b>To establish a preliminary foundation for developing a serum proteomics diagnostic model of nasopharyngeal carcinoma (NPC) by comparing the differences in serum protein fingerprints between patients with NPC and healthy subjects and between different types of NPC.</p><p><b>METHODS</b>The serum samples of 41 patients with different types of NPC and 20 healthy subjects were collected. Surface-enhanced laser desorption ionization time-of-flight mass spectrometry (SELDI-TOF-MS) were used to detect the blood samples to obtain serum protein mass spectrum, i.e. serum protein fingerprinting. Biomarker Wizard and Biomarker Patterns system software were used to compare the differences in serum protein mass spectrum between NPC patients and healthy subjects and between different types of NPC, and to screen out the NPC-related serum proteins.</p><p><b>RESULTS</b>Compared with the healthy control, NPC patients emerged 9 very prominent protein peaks (P < 0.001), with the combined differential peaks. No significant difference was found in relative amount of serum proteins with different molecular mass between different types of NPC (P > 0.05).</p><p><b>CONCLUSIONS</b>The serum marker proteins and specific protein fingerprints of NPC can be screened out by SELDI-TOF-MS, which could be used to develop a serum proteomics model for clinical screening and early diagnosis of NPC.</p>

ABSTRACT

<p><b>BACKGROUND</b>The initiation and expansion of China's national free antiretroviral therapy program has led to significant improvement of survival among its participants. Success of further scaling up treatment coverage rests upon intensifying HIV screening and efficient linkage of care. Timely CD4 cell count testing after HIV diagnosis is necessary to determine whether a patient meets criteria for antiretroviral treatment, and represents a crucial link to engage HIV-infected patients in appropriate care, which has not been evaluated in China.</p><p><b>METHODS</b>We evaluated all patients ≥ 16 years who tested HIV positive from 2005 to 2009 in Yunnan and Guangxi. Multivariate Logistic regression models were applied to identify factors associated with lack of CD4 cell count testing within 6 months after HIV diagnosis.</p><p><b>RESULTS</b>A total of 83 556 patients were included. Over the study period, 30 635 (37%) of subjects received a CD4 cell count within 6 months of receiving the HIV diagnosis. The rate of CD4 cell count testing within 6 months of HIV diagnosis increased significantly from 7% in 2005 to 62% in 2009. Besides the earlier years of HIV diagnosis, negative predictors for CD4 cell count testing in multivariate analyses included older age, not married or unclear marriage status, incarceration, diagnosis at sexual transmitted disease clinics, mode of HIV transmission classified as men who have sex with men, intravenous drug users or transmission route unclear, while minority ethnicity, receipt of high school or higher education, diagnosis at voluntary counseling and testing clinics, and having HIV positive parents were protective.</p><p><b>CONCLUSIONS</b>Significant progress has been made in increasing CD4 testing among newly diagnosed HIV positive patients in Yunnan and Guangxi from 2005 - 2009. However, a sizable proportion of HIV positive patients still lack CD4 testing within 6 months of diagnosis. Improving CD4 testing, particularly among patients with identified risk factors, is essential to link patients with ART services and optimize treatment coverage.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the infection of Cryptosporidium and its epidemiological characteristics in AIDS patients of Southern China.</p><p><b>METHODS</b>Stool samples colleted from AIDS confirmed patients. The samples were detected for oocyst of Cryptosporidium by acid fast bacteria stain and indirect fluorescent antibody stain respectively, CD4 count was detected by Flow Cytometry.</p><p><b>RESULTS</b>212 samples of fresh stool obtained from the AIDS patients who live in Guangdong and Yunnan province. The total infection rate of Cryptosporidium in AIDS patients was 4.25% (9/212), the infectious rate of oocyst in the group of 50- 59-years-old was significantly higher than those in 30-39 (P < 0.01); the infectious rate of oocyst in patients with antiretroviral therapy (ART) was also significantly lower (P = 0.0000); we found the patients coinfected with Cryptosporidium with CD4 count all below 100 cells/microl. However, there were no any difference between the infectious rate to the patient's gender, areas and stool shape.</p><p><b>CONCLUSION</b>AIDS patients infected by Cryptosporidium are not rare in southern China, and the infectious rate was lower than western country. Patients received ART could decrease the infectious rate of Cryptosporidium, Cryptosporidium always happen in patient whose CD4 count was very low (< 100 cells/microl).</p>

ABSTRACT

<p><b>OBJECTIVE</b>To search for an effective therapy for allergic rhinitis.</p><p><b>METHODS</b>One hundred and twenty cases of allergic rhinitis were randomly divided into a treatment group (n=60) treated mainly with penetration needling, Yintang (EX-HN 3)-through-Bigen (root of nose), Sibai (ST 2)-through-Bigen, Yingxiang (LI 20)-through-Bigen, and a control group (n=60) treated with oral Biyankang, 4 tablets each time, 3 times daily. Their therapeutic effects were compared.</p><p><b>RESULTS</b>The total effective rate was 85.0% in the treatment group and 60.0% in the control group, with a very significant difference between the two groups (P<0.01).</p><p><b>CONCLUSION</b>The penetration needling as main therapy has a definite therapeutic effect on allergic rhinitis, which is significantly better than that of oral Biyankang.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To screen the sensitive chemotherapeutic agents to human endometrial carcinoma cell line-1 (HECCL-1) and study its mechanism.</p><p><b>METHODS</b>MTT method was used to examine the relative inhibition ratios (RIRs) of various concentrations of 18 chemotherapeutic agents to HECCL-1. Cell cycle, apoptosis and expression of MDR1 protein were detected by FCM.</p><p><b>RESULTS</b>Nine of the chemotherapeutic agents studied obviously inhibited the proliferative activity of HECCL-1 in a dose-dependent manner. The order of sensitivity was as follows: adriamycin (ADM), oxaliplatin (L-OHP), carboplatin (CBP), cisplatin (DDP), taxol (TAL), epirubicin (EPI), mitoxantrone (MIT), dactomycin (ACTD) and 5-fluorouracil (5-Fu). FCM showed these agents could significantly reduce the proportion of cells in G0-G1 phase, and increase the proportion of cells in S and G2-M phase (P < 0.05). Cell apoptosis was observed in 11 chemotherapeutic agents at their peak concentration. MDR expression was induced after using EPI, 5-Fu, hydroxycamptothecin (HCPT) and MIT.</p><p><b>CONCLUSION</b>HECCL-1 is sensitive to a number of the chemotherapeutic agents studied. Induced apoptosis may be the major mechanism of drug sensitivity, and acquired drug-resistance may be the critical reason against continued administration.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the compatibility of composite herbal medicines of the Ling Gui Zhu Gan Decoction.</p><p><b>METHOD</b>Ethanol extract test solutions of the different combinations were prepared according to the orthogonal layout L16(4(5)). Pharmacologic experiments, such as the time of surviving of mice in shortage of oxygen in regular air pressure, the antagonizing effect on arrhythmia induced by chloroform and diuresis were carried out with the solutions. Variance analysis, canonical correlation and stepwise regression analysis were applied to interrelate the amount of each drug and the pharmacologic data.</p><p><b>RESULT</b>The results confirmed that Fuling and Guizhi are the basis, while Baizhuand Gancao are the adjuvans, which is conformed to the theory of TCM.</p><p><b>CONCLUSION</b>This study provides a significant try for studying the compatibility of composite herbal medicines.</p>

ABSTRACT

Objective To evaluate the value of positron emission tomography(PET)-CT imaging combined with continual detection of CA_(125)in serum for diagnosis of early recurrent ovarian epithelial carcinoma.Methods Twenty six patients received PET-CT imaging,who were all diagnosed as primary epithelial ovarian cancer of stage Ⅱ-Ⅳ and had complete remission after cytoreductive surgery and multiple courses of chemotherapy in Shandong Provincial Cancer Hospital.After a steady period,all patients experienced progressive rising of CA_(125)values 3 times in 2 months.But no positive lesion was found by CT, or although suspicious positive focus was found,the recurrent and(or)metastatic extent was not definite. Out of 26 patients,16 were delivered rechemotherapy and(or)radiotherapy,and 10 received re- cytoreductive surgery.Results(1)Of 26 patients,the value of CA_(125)was more than 35 kU/L in 17,and in 14 of 17,pelvic or abdominal cavity recurrence was diagnosed by CT and PET-CT,and 4 showed simuhaneously distant metastasis on PET-CT.In the remaining 3 patients of which CT findings were negative,2 had pelvic and abdominal cavity recurrence,and one had bone metastasis on PET-CT.Of 9 patients with progressive rising CA_(125)levels but the value was less than cut-off(
